The Science Behind Paternity Testing
A paternity DNA test works by comparing specific locations in the DNA of a child with those of an alleged father. Every person inherits half their DNA from their biological mother and half from their biological father. By examining a set of genetic markers called STRs (Short Tandem Repeats), labs can determine with extremely high probability whether a biological relationship exists.
How the Test Is Conducted
- Sample collection: A simple cheek swab (buccal swab) is used to collect DNA from the inside of the cheek. It's painless, non-invasive, and can be done at home or in a lab setting.
- DNA extraction: The lab extracts DNA from the cells collected on the swab.
- Marker analysis: The lab examines typically 16–24 STR loci (locations on chromosomes) from both the child and the alleged father.
- Comparison: If the alleged father's markers match what would be expected for a biological father at each locus, paternity is supported. Mismatches at multiple loci indicate the person is not the biological father.
- Statistical calculation: Results are expressed as a Probability of Paternity — a figure above 99.9% is considered a positive inclusion; 0% is a definitive exclusion.
Accuracy: What the Numbers Mean
Modern paternity tests are extraordinarily accurate. A result of 99.9% or higher is the accepted threshold for confirming paternity in most jurisdictions. A 0% result is a definitive exclusion — meaning the tested individual is definitively not the biological father.
It's important to understand that 99.9% is not 100% — but reaching absolute 100% certainty is statistically impossible. In practice, modern STR-based testing is considered definitive for legal and personal purposes.
Types of Paternity Tests
At-Home Paternity Tests
These kits are mailed to you, samples are collected at home, and results are typically delivered online within a few days. They are best for personal peace of mind and are generally not admissible in court because there is no verifiable chain of custody for the samples.
Legal Paternity Tests
For use in legal proceedings — such as child support, custody disputes, inheritance, or immigration cases — a legal (chain-of-custody) paternity test is required. Key differences include:
- Samples must be collected by a neutral third party (e.g., a medical professional or authorized collector)
- All participants must provide photo ID
- Samples are sealed and documented to maintain an unbroken chain of custody
- Results are sent directly to the requesting party or court
Prenatal Paternity Tests
It is possible to determine paternity before birth using either:
- Non-Invasive Prenatal Paternity (NIPP): A blood sample from the mother is analyzed for fetal DNA fragments — safe and available from around 7–8 weeks of pregnancy.
- Amniocentesis or CVS: Invasive procedures that carry small risks — typically only considered when medically necessary for other reasons.
Beyond Paternity: Other Relationship Tests
DNA testing can confirm many other biological relationships beyond father and child:
- Maternity testing: Confirms a biological mother-child relationship (sometimes needed in adoption or surrogacy cases)
- Sibling testing: Determines whether two individuals share one or both biological parents
- Grandparentage testing: Used when the alleged father is unavailable
- Aunt/Uncle testing: Establishes biological relationships through the paternal family
